PhD position in functional characterization of plant pathogen effector proteins

Vascular Xanthomonas pathogens have a distinct infection strategy compared to other bacterial pathogens, as they infect their host via hydathodes, which are water pores at the leaf margin that are connected to the xylem endings. This provides the pathogen access to the vasculature for systemic spreading through the leaf. During different stages of the infection process Xanthomonas injects effector proteins into host cells to interfere with plant physiology and immune signaling thereby promoting bacterial proliferation. The overall goal in this project is to identify and characterize key plant processes manipulated by Xanthomonas pathogens. The approach will be to uncover the interactions between pathogen effectors and their target host proteins. Identifying host targets will pinpoint the genes and/or pathways that function as susceptibility factors, as the pathogen relies on them for successful infection. We will then genetically engineer plants modifying those effector targets and test resistance against Xanthomonas.

First, you will screen for host proteins manipulated by Xanthomonas effectors using a readily available protein proximity labelling technique and mass spectrometry-guided protein identification. Subsequently, you will characterize the function of the identified plant proteins during infection. For this, we will make use of the molecular tools available in the model plant Arabidopsis thaliana, which belongs to the same family as Brassica crops. This involves engineering plant knockouts using established CRISPR-Cas technology and applying advanced transcriptomics experiments. In particular, you will investigate the effector function during early infection stages in hydathodes. Lastly, you will test the function of the identified gene in Brassica plants during Xanthomonas infection.

Tasks and responsibilities:

• complete a PhD thesis within the official appointment duration of four years;
• perform your experiments in a systematic and controlled manner and keep records by properly documenting and

organizing your work;

• be an active member of the research group and take responsibility for shared tasks;
• discuss your work with group members and during company update meetings. Incorporate feedback and give

input to others.

• take a leading role in writing manuscripts for publication in peer-reviewed journals;
• participate in the PhD training programme of the University of Amsterdam and the Dutch Experimental School

of Plant Sciences;

• assist in teaching and supervising Bachelor and Master internships and tutoring students.

Docenten Psychobiologie

Je gaat over het algemeen de volgende werkzaamheden uitvoeren:

• geven van diverse practica en werkgroepen in alle studiejaren en samen met het begeleidingsteam hiervoor inwerken
• geven van feedback en beoordelen van opdrachten (Nederlands en Engels);
• geven van individuele begeleiding;
• deelnemen aan docentenoverleg;
• fungeren als mentor voor de eerstejaars studenten: je hebt mentorgesprekken met studenten en verwijst hen door naar de studieadviseur indien nodig.
• surveilleren bij tentamens en essayvragen nakijken.

Je geeft in ieder geval werkgroepen voor Academische Basisvaardigheden. De werkgroepen worden wekelijks gezamenlijk voorbesproken. De overige werkgroepen en practica zullen op basis van je expertisegebied in combinatie met die van andere docenten worden samengesteld.

Voorbeelden van overige practica zijn: Jaar 1: Genetica en Evolutie, Cognitieve Psychologie, Celbiologie, Neurofysiologie, Neuroanatomie. Jaar 2: Experimentele Psychologie, EEG, Programmeren (Python), Neurofarmacologie, Moleculaire Celbiologie.

Voorbeelden van individuele begeleiding zijn: Review jaar 2 (jaar 2), docenten met een PhD kunnen optreden als 2e beoordelaar bij de bachelorprojecten (jaar 3).

Promovendus Groeigerichte Overtuigingen en Handelen van Leraren in een Selectief Onderwijsstelsel

Als promovendus onderzoek je groeigerichte overtuigingen en handelingen van leraren in verschillende VO-scholen. Door te kijken naar categorale en brede scholen, onderzoek je of de mate van selectiviteit samenhangt met verschillen in het denken en handelen van leraren. Vervolgens ga je aan de slag met het ontwikkelen van een interventie gericht op het versterken van groeigerichte overtuigingen en handelen. In nauwe samenwerking met laatstejaars studenten van de tweedegraads lerarenopleidingen Nederlands en Wiskunde ontwikkel je interventiematerialen en realiseer jij de implementatie en evaluatie ervan. Je onderzoek resulteert in een academisch proefschrift, bestaande uit vier wetenschappelijke artikelen. Je participeert in de Community of Practice. Daarnaast maak je deel uit van het onderzoeksteam waarvan ook de promovendus van het het PO-gerichte project – die wordt aangesteld bij de afdeling Sociologie – en alle betrokken begeleiders deel uitmaken. In verband met de nauwe samenwerking met de lerarenopleidingen van de HvA in dit project, behoort het inrichten van een werkplek op de HvA tot de mogelijkheden.

Junior Onderzoeker Forensische Orthopedagogiek

Je voert binnen dit project drie deelstudies uit:

1. ontwikkeling en implementatie van een registratiesysteem en infrastructuur om uitkomsten van de jeugdbescherming te monitoren, en van data te leren;
2. onderzoek naar effecten van een ondertoezichtstelling op jeugdigen en hun ouders;
3. single case study waarin je gezinnen met een OTS intensief volgt gedurende 1,5 jaar.

Dit project wordt uitgevoerd in samenwerking met professionals uit de praktijk.

Je bent verantwoordelijk voor de uitvoering, werving, en rapportage van het onderzoek, en onderhoudt hiervoor nauw contact met professionals en gezinnen.
